The viability of the Biuret method for protein determination in cheese: a comparative study

Abstract

The determination of proteins in foods is important both for nutritional evaluation and for ensuring the quality standards of the final product. The official method, Kjeldahl, is accurate but time-consuming. Therefore, the objective of this study was to evaluate the Biuret method as a simpler, faster, and more economical alternative for the determination of proteins in cheeses. Three types of cheese (Camembert, Ricotta, and Tomme Negro D’Oeste) were analyzed using both methods. To evaluate the method, accuracy and precision were determined, as well as preliminary tests with different solvents for protein extraction from the cheeses. Among the solvents tested, 0.5 M sodium hydroxide showed the best performance. Accuracy was higher in Ricotta (73.4%), followed by Camembert (35.0%) and Tomme Negro D’Oeste (29.4%). All matrices exhibited good precision (relative standard deviation < 10%). Protein extraction from cheese using 0.5 M NaOH, shaking at 40°C and 170 rpm for 15 minutes, and vortexing for 1 minute was the most efficient; however, it still showed low recovery of cheese proteins. Therefore, the determination of proteins in cheeses using the Biuret colorimetric method is not a suitable alternative to replace the official method.
